 AB    OBJECTIVE: Immunofluorescence assay (IFA) which is capable of
       simultaneously detecting antibodies against closely related HIV-1 and
       HIV-2 was established and evaluated for the ability to serologically
       discriminate HIV-1 and HIV-2 infections. METHODS: IFA employing three
       types of cells, HIV-1-infected (U937/LAI), HIV-2-infected
       (Multinucleated giant cells which were formed by cocultivation of
       MOLT-4#8 and MOLT-4/GH-1) and uninfected (HeLa) cells whose morphology
       varies within one field of microscopy was performed. Serum specimens
       from 95 HIV-1-infected, 26 HIV-2-infected and 3 HIV-1/HIV-2
       dually-infected individuals and 106 seronegative blood donors were
       tested using above mentioned cells. RESULTS: All the samples from
       individuals infected with HIV were confirmed to contain antibodies to
       HIV-1 and/or HIV-2. None of the sera from blood donors were positive.
       Serum specimens from HIV-1-infected or HIV-2-infected individuals were
       diagnosed as infections with HIV-1 (85/95) and HIV-2 (22/26),
       respectively. Other 14 (10/95 and 4/26) were shown to be seropositive
       for both HIV-1-infected and HIV-2-infected cells. DISCUSSION AND
       CONCLUSION: Our results of newly-developed IFA was capable of
       distinguishing most (88%) of HIV-1 and HIV-2 infections at one time.
       This assay appears to be useful for screening as well as for confirming
       of both HIV-1 and HIV-2 infections simultaneously.
