AB OBJECTIVE: The establishment of a simple and sensitive method for the
multiple detection of antibodies to HIV-1 and HBs that would eliminate
tedious procedures and elaborate instrumentation. METHOD: This method is
based upon chemiluminescent immunochromatography using glassfiber strip
and instant film for photo detection. The test strip is shown in Fig. 1.
After dropping 50 microliters of sample on the sample zone, the
immunoreaction is started by the developing solution, and the following
enzyme reaction is carried out with CSPD as chemiluminescent substrate.
The strip is developed and the enzyme is reacted for 30 minutes. The
strip is then exposed on the instant film for 30 seconds. RESULTS: Low
titer sample with 2(5) dilution positive by particle agglutination (PA)
test showed positive result on the film. In this method, 15 positive
samples in the PA test showed positive result, but 5 negative samples in
the PA test did not react. DISCUSSION AND CONCLUSION: In the present
method, we were able to achieve a simple and sensitive assay with
results easily read by naked eye. Moreover, with this method the
simultaneous detection of antibodies to both HIV-1 and HBs for primary
blood screening is now possible. Assay conditions still need to be
improved to obtain more rapid assay with shortened reaction time.
