AB OBJECTIVE: AMPLICOR HIV-1 kits, which had been developed as an HIV-1
provirus detection test by PCR method, have been evaluated for its
clinical diagnostic application. We have compared AMPLICOR HIV-1 with
virus isolation and HIV-1 antibody test. METHODS: (1) HIV-1 antibody
test (Particle agglutination and Western Blot), (2) Virus isolation, (3)
PCR by AMPLICOR HIV-1 test kit. SAMPLES: Two hundred sixty-two of HIV-1
antibody positive and 67 of HIV-1 negative blood samples derived from
hemophiliacs, who had received blood products. Three of new born infants
from HIV-1 sero-positive mothers. RESULTS: The results from PCR were
consistent with those from antibody test and clinical aspects.
Thirty-nine of HIV-1 antibody positive samples have been tested by PCR
and virus isolation. Thirty-nine of 39 (100%) were positive by PCR and
12 of 39 (30.8%) were positive by virus isolation. One of three new born
infants from HIV-1 positive mothers were known HIV-1 infection by PCR.
We did not see the silent infection in hemophiliacs. CONCLUSIONS:
AMPLICOR HIV-1 is considered as useful clinical diagnostic for HIV-1
proviral DNA detection and it suggested that the kit would be useful for
diagnosis of infants from sero-positive mothers.
