AB OBJECTIVE: Most of AIDS-related lymphomas (AIDS-ML) are infected with
EBV. In order to investigate the origin of these neoplastic cells, we
made an analysis using molecular biologic methods. METHOD: We examined
35 cases of HIV-infected lymphnodes that were in PGL stage. For the
demonstration of EBERs, we used in situ hybridization, and for the
lymphocyte markers and EBV-produced oncoprotein LMP-1 (latent membrane
protein 1). we used immunostaining method. 20 cases of HIV-negative
lymphnodes were also studied as controls. RESULTS: EBV-encoded small
RNAs (EBERs)-positive cells were found in 15 out of 35 cases. All 20
cases of controls were negative for EBER. Immunostaining revealed many
of the EBER-positive cells to be positive for LMP, and most of the
EBER-positive cells to be in B cell lineage with CD20 and the remaining
part were macrophage positive for CD68. DISCUSSION: We found that much
increased number of EBV-bearing B cells, (and macrophages) are present
in lymphnodes of HIV-infected patients. We believe that these
EBV-positive B cells expressing EBERs and LMP-1 are the very precursor
       cells of AIDS-lymphomas.
